Bacterial Transformation of Vibrio Fischeri's Bioluminescent Plasmid onto Escherichia Coli

The purpose of this experiment was to test factors that would influence the transformation of a bioluminescent plasmid from Vibrio fischeri onto Escherichia coli DNA. The factors that were tested were the addition of ampicillin to agar, the salinity of agar and temperature for E.coli incubation. The experiment was performed over thee weeks, during which time we observed the uptake of the luminescent plasmid by observing luminescent colonies of E.coli. We found what factor(s) had the greatest effect on the luminescence of E.coli.
